1. Before electrophoresis, it is forbidden to connect the power cord attached to the electrophoresis tank to the power supply of the electrophoresis instrument.

2. Assemble the electrophoresis tank:

1) Insert the four fixing screws into the corresponding holes of the liquid storage tank with the red electrode, and then put the lower tank on the table.

2) Hold the grooved rubber mold frame with your left hand, hold the edges of both sides of the flat glass with your right hand and insert it into the lower groove of the rubber mold frame, and then insert the glass plate with the glass spacer into the corresponding upper groove in the same way Inside.

3) Put the rubber mold frame with glass flat on the upside-down liquid storage tank frame, pay attention to make the side with the glass strip in contact with the lower tank, so that the lower edge needs to be aligned with the lower edge of the liquid storage tank, and make the The protruding arc on the rubber frame is aligned with the center of the plexiglass of the reservoir.

4) Take another liquid storage tank with black electrode with both hands and match it with the lower tank placed on the table, so that the protruding arc on the rubber frame is located in the center of the plexiglass of the upper liquid storage tank.

5) Install four nuts, erect the electrophoresis tank vertically, put it on the table, and then tighten the bolts, so that the four bolts evenly stressed. Use 10% agar sealant between the glass plate at the lower end of the chamber and the rubber frame gap.

6) Install rubber hoses at the two water outlets of the upper and lower tanks, and clamp the two water outlets with clips to make them airtight.

3. Glue filling: Add the separation glue into the middle of the glue chamber formed by the two glass plates, stop pour the concentrated glue after solidification, and insert the sample grid at the same time. To prevent leakage, distilled water should be added to the upper and lower reservoirs.

4. Add samples: loosen the clamps on the two rubber tubes connecting the upper and lower storage tanks, let go of the distilled water in the tanks, and tighten the clamps after the water flows Then add buffer solution to the upper and lower storage tanks, Then unplug the sample grid and add the sample into the sample grid.

5. Electrophoresis: Connect the upper tank electrophoresis wire to the negative pole of the electrophoresis instrument, connect the lower tank to the positive pole of the electrophoresis instrument, connect the cooling water, and connect the electrophoresis instrument after checking that it is correct. Prohibit people from contacting with liquid during electrophoresis.

6. End electrophoresis: After the electrophoresis experiment is over, first turn off the electrophoresis instrument, then unplug the electrophoresis wire, cut off the cooling water, let go of finally, loosen the bolts, remove the glue chamber, and place it in the gap at the lower corner of the two glass plates. Gently pry with the back of the razor blade to separate the adhesive surface from a glass plate.
