In the past, the stretching tool for making paraffin sections was : a paraffin section stretcher . Although there are ready-made ones on the market , the price is generally around 300 yuan . For infrequent users , it is undoubtedly an additional expense . Especially for units with less scientific research funds such as normal colleges , technical secondary schools , and middle schools , there are indeed difficulties in purchasing them .
In my work, I experimented with a Water Bath instead of a paraffin section stretcher to make paraffin sections , and achieved satisfactory results . The method is as follows :
According to伸展器烫板要求的35℃的温度,将水浴锅的温度调节旋扭调到50℃。因为水浴锅中的温度要高于其外面的温度。将水浴锅的盖盖好,再放上一块与水浴锅盖面积大小相等的玻璃,将粘贴好材料的载玻片平放到玻璃上,此时,蜡片因受热而伸展摊平,达到了与石蜡切片伸展器烫板同样展片的目的。实践证明:使用水浴锅为石蜡切片伸展器是切实可行的。用这种方法制成的切片与用石蜡切片伸展器制成的切片相比较,其质量效果是一致的。但是使用水浴锅为伸展器应注意以下几个问题:
1. The temperature adjustment of the Water Bath depends on the season and room temperature . Summer (high room temperature) can be slightly lower , around 40°C ; winter (low room temperature) can be slightly higher , around 50 °C .
2. A thermometer should be placed on the lid of the Water Bath , and the temperature of the Water Bath should be adjusted up or down according to the thermometer . Keep its thermometer at about 35 ℃ , which is conducive to spreading .
3. You can also boil the water in the Water Bath first , and then lower it to 50 ℃ , which can save the preheating time .
